Experimental details

Western blot analysis of OLIG2 in mouse brain lysate. Samples were incubated in OLIG2 polyclonal antibody (Product # PA5-23456) using a dilution of 2 µg/mL followed by an anti-rabbit HRP secondary antibody. Separated on a 12% gel using SDS-PAGE, protein transferred to PVDF membrane and blocked in 5% milk-TBST. Detection: chemiluminescence.

Experimental details

Western blot was performed using Anti-OLIG2 Polyclonal Antibody (Product # PA5-23456) and a 35 kDa band corresponding to OLIG2 was observed in the tissues tested. Tissue extracts (30 µg lysate) of Mouse Pup Brain (Lane 1), Mouse Brain (Lane 2) and Mouse Liver (Lane 3) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

Experimental details

Immunocytochemistry analysis of OLIG2 in Neuro2a cells fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. Samples were incubated in OLIG2 polyclonal antibody (Product # PA5-23456) using a dilution of 1:200 dilution overnight at 4 °C followed by DyLight 488 (Green). Alpha tubulin was used as a co-stain at a 1:1000 dilution and detected with DyLight 550 (Red). Nuclei were detected with DAPI (Blue). Cells were imaged using a 40X objective.
