- Western blot analysis was performed on membrane enriched cell extracts (30 µg lysate) of Molt-4 (Lane 1), Jurkat (Lane 2), Raji (Lane 3), Ramos (Lane 4), THP-1 (Lane 5) and U-937 (Lane 6). The blot was probed with Anti- CD38 Monoclonal Antibody (Product # MA5-15570, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/mL, 1:4000 dilution). A 45 kDa band corresponding to CD38 was observed across all the cell lines tested except THP-1 and U-937.

- Western blot was performed using CD38 Monoclonal Antibody (6E12D) (Product # MA5-15570) and a ~46 kDa band corresponding to CD38 was observed in Ramos, Daudi, A549, MOLT-4, Jurkat, Raji . Whole cell extracts (30 µg lysate) of Ramos (Lane 1), Daudi (Lane 2), A549 (Lane 3), MOLT-4 (Lane 4), Jurkat (Lane 5), Raji (Lane 6), THP-1 (Lane 7), H1299 (Lane 8), HCT 116 (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
